Abstract:The research used pigskin as raw material, which was treated by enzymes, to extract collagen. To confirm and optimize the best extraction condition and process, the structural properties of collagen product were characterized. The experiment results showed that the extraction yield of collagen product was 65.33 % and the purity was 87.38 %. The thermal denaturation temperature was 118.79 ℃. The maximum UV absorption wavelength was 203 nm. The relative molecular mass was 3.63 ×105 g/mol. Infrared spectrum and round two chromatographic experiments showed that the product had a typical collagen absorption peak, indicating the three helical structure of collagen extracted was intact; SDS -PAGE gel electrophoresis test showed that the product was mainly composed of three sub components.
