响应面法优化厚壳贻贝多糖提取工艺及抗肿瘤活性研究

中文摘要: 以厚壳贻贝多糖提取率为响应值,在单因素试验的基础上,以液料比、酶解温度、酶解时间和 pH 值为试验因 素,采用响应面法建立数学模型,筛选最佳提取工艺条件;建立小鼠 S180 实体瘤模型,分为模型对照组、厚壳贻贝多 糖实验低、中、高剂量组、环磷酰胺对照组。测定小鼠肿瘤生长抑制率、脾脏指数和胸腺指数。通过二次回归模型响应 面分析,确定厚壳贻贝多糖的最佳提取工艺条件为液料比 9.8 mL/g、酶解温度 46 ℃、酶解时间 133 min 和 pH5.9,在此 条件下厚壳贻贝多糖提取率为 19.2 %,与模拟方程得出的 18.9 %无显著性差异;并且中、高剂量厚壳贻贝多糖对小鼠 的肿瘤抑制率分别为 36.8 %和 51.9 %。得到酶解法提取厚壳贻贝多糖的最佳提取工艺并证实其免疫增强作用。

Abstract:Employing a three-level and four-variable Box-Behnken design and setting the Mytilus coruscus polysacchrides(MCP)extraction yield(%)as response value, the optimum enzymatic extraction conditions were as follows: extraction time was 133 min, extraction temperature was 46 ℃, extraction pH was 5.9, and ratio of liquid to fresh flesh was 9.8 mL/g. Under the optimized conditions, the predict extraction yield indicated from the RSM model was 18.9 %, which showed compliance to the experimental yield(19.2 %). The antitumor activity was investigated in mice. The tumor growth inhibitory rate, spleen index and thymus index were calculated. The tumor inhibition rates of medium and high dose MCP were 36.8 % and 51.9 %, respectively, which were significantly different with model group. Spleen index and thymus index significantly increased in medium and high dose MCP group. It was obviously that MCP could improve the immunity and inhibit the growth of tumor.
