中文摘要: 以安溪铁观音为原料,以产品得率、茶多糖含量和 DPPH 自由基清除率为评价指标,考察先 70 %醇提+后水提 和先水提+后 70 %醇沉等 2 种提取方式对茶多糖特性的影响。结果表明,与先水提+后 70 %醇沉相比,采用先 70 %醇 提+后水提制备的粗茶多糖得率低,而多糖含量及 DPPH 自由基清除率则极显著提高。进一步通过正交试验分析发现, 料液比和温度对粗茶多糖的 DPPH 自由基清除率呈显著性影响,料液比对粗茶多糖含量呈显著性影响,乙醇浓度则呈显著 性影响粗茶多糖产品得率。为获得高活性且含量高的粗茶多糖,最优化工艺参数为乙醇浓度 60 %、料液比 1 ∶ 10(g/mL)、提 取温度 90 ℃和提取时间 20 min。

Abstract:Anxi Tieguanyin was employed to assess the 2 extraction methods of pre-70 % ethanol extracting + post-water extracting and pre-water extracting + post-70 % ethanol precipitating via crude tea polysaccharide yield, crude tea polysaccharide content and DPPH·scavenging. The results showed that crude tea polysaccha- rides by pre-70 % ethanol extracting + post-water extracting had a lower yield, significantly higher content and more stronger DPPH·scavenging than pre-water extracting + post-70 % ethanol precipitating. For the further study by orthogonal test, found that the influences were significantly exhibited by the ratio of solid to liquid and temperature for DPPH·scavenging, the ratio of solid to liquid for the content of crude tea polysaccharides and ethanol concentration for the yield of crude tea polysaccharides. The optimal extracting conditions of crude tea polysaccharides with high bioactivity were as follows: 10 time liquid of solid, extracted at 90 ℃ for 20 min with 60 % ethanol.
