中文摘要: 本试验以蛋白质脱除率、多糖保留率以及两因素的综合指标为评价指标,采用 Sevage 法、盐酸-乙醇等电点法 和 TCA 法,分析比较其对平菇多糖脱蛋白效果的影响。试验结果表明,平菇多糖中蛋白质脱除率最高的为 Sevage 法,其 次为 TCA 法;而多糖保留率最高的为盐酸-乙醇等电点法,其次为 TCA。综合考虑两因素的影响,以 TCA 法效果最好。 利用 Box-Behnken 试验设计,获得 TCA 法脱蛋白工艺的回归方程,再经响应面分析,得出脱蛋白最佳方案,即:TCA 浓 度 5.6 %,处理时间 33 min,处理温度 48 ℃,此时其综合评价平均值为 83.96 %,蛋白质脱除率为 89.5 %,多糖保留率为
78.4 %。
Abstract:The effects of protein removal from polysaccharide of Pleurotus ostreatus were analyzed by comparing the three methods of Sevage, hydrochloric acid-ethanol isoelectric point and TCA with protein removal rate, polysaccharide retention rate and comprehensive index as evaluation index. The result showed that protein re- moval rate of polysaccharide by the method of Sevage was the highest, and followed by the method of TCA. The best way for polysaccharide retention rate was the method of hydrochloric acid-ethanol isoelectric point, and second was TCA. And in consequence the TCA method was the most effective one among the three ways by con- sidering the two factors of protein removal rate and polysaccharide retention rate. A regression equation of the protein removal from polysaccharide of Pleurotus ostreatus by the way of TCA was obtained using the Box -Behnken test design, and then the optimal conditions of the process for protein removal was established by re- sponse surface analysis, which was TCA concentration 5.6 %, processing time 33 min, and temperature 48 ℃. Under these conditions, the average of comprehensive evaluation was 83.96 %, the protein removal rate and polysaccharides retention rate was 89.5 % and 78.4 %, respectively.
