中文摘要: 研究大孔吸附树脂分离纯化菜芙蓉黄酮的最佳工艺条件。以总黄酮吸附量和解吸量为指标,进行静态吸附和解吸试验对14种型号大孔树脂进行筛选,再通过动态吸附和解吸试验对纯化工艺参数进行优化。Z801 大孔树脂对菜芙蓉总黄酮的吸附与解吸性能最佳。HZ801纯化菜芙蓉黄酮的最佳工艺条件为:上样浓度为1 mg/mL,上样流速2 mL/min,上样量为140 mL;依次用2 BV水洗脱,用70 %乙醇以2 mL/min的速率洗脱2.2 BV。在优化工艺条件下,菜芙蓉黄酮的平均吸附率是95.03 %,纯化倍数 4.04。HZ801型大孔树脂富集黄酮的效果最佳,是一种较理想的分离纯化介质。
Abstract:The optimum conditions of purification of total flavones in Hibiscus manihot flower were investigated by using the macroporous adsorption resins. The the amount of total flavonoids by adsorption and desorption as an index,14 kinds of macroporous resins were screened by static adsorption and desorption experiments and then purification process parameters were optimized by dynamic adsorption and desorption tests. HZ801 resin was had the best adsorption and desorption effects. The optimal purification conditions are as follows:the sample concentration is 1 mg/mL ,the sample flow rate is 2 mL/min ,
the sample size is 140 mL;the sampleis elutedby water with 2 BV and 70 % ethanol of 2.2 BV,the eluting velocityis 2 mL/min. In the optimal conditions, theaverage adsorption rate of total flavones in Hibiscus manihot flower was 95.03 %,and the purification factor is 4.04. HZ801 macroporous resin has the best effect on the enrichment of flavonoids,It is an ideal separation andpurification medium.
