中文摘要: 采用液液萃取法从甘蔗梢粗黄酮中纯化黄酮成分,单因素试验分别考察萃取剂、萃取温度、萃取pH 值和萃取体积比对黄酮成分纯化效果的影响。在单因素试验的基础上,根据CCD 中心组合设计原理对试验数据进行响应面 分析,建立萃取液中黄酮含量的多元二次回归方程,得到优化组合条件。结果表明,液液萃取法从甘蔗梢粗黄酮中纯 化黄酮成分较优的工艺条件为:萃取剂为正丁醇,萃取pH 值为 3.27,萃取体积比为 3: 1,萃取温度为 50℃,此时相对于甘蔗梢的黄酮得率为166.4 mg/kg,与模型预测值相近。

Abstract:Response surface methodology was employed to optimize the purification of flavonoids from the crude flavonoids extracts of sugarcane tips. The influence of three purification variables on flavonoids content was investigated ,including extraction pH value,extraction volume ratio and temperature. A central composite design was used to investigate the best conditions for purification of flavonoids produced by sugarcane tips,and a multivariable quadratic regression equation was developed. The results showed that the optimal purification conditions about flavonoids from the extracts of sugarcane tips were as follows:the sugarcane tip extractants were dissolved at 50 ℃in n-butanol,pH value = 3.27,extraction volume ratio was 3:1. Under above-mentioned con原ditions,the content of flavonoids was up to 166.4 mg/kg,which was similar to the predicted value.
