中文摘要: 为了研究葛根多糖的基本理化特性,采用脱脂脱蛋白后的葛根多糖过DEAE-52纤维素柱得到的样品PS-D1、PS-D2、PS-D3为原料,研究3组样品的分子量分布,基本外貌形态,刚果红反应和体外抗氧化的测定。结果表明:3种组分的分子量分散系数比较集中,分别为1.4、1.9和1.4,外貌形态经SEM扫描分析为纤维树枝状,片状或碎屑状堆积,刚果红分析表明3种多糖不具有三螺旋结构,在水溶液中表现为无规线团链构象。体外抗氧化结果表明葛根多糖有较好的DPPH自由基的清除能力且它们的半抑制率IC50值分别为3.14、2.85、2.31 mg/mL,并且均含有一定的清除ABTS+自由基的能力,其中PS-D3的等价抗氧化能力(TEAC)最好,为(53.83 μmol Trolox/g样品)。葛根多糖的基本理化特性研究为葛根的开发及综合利用提供了理论参考。
Abstract:In order to study the basic physical and chemical properties of the polysaccharides from Pueraria,the raw material PS-D1,PS-D2 and PS-D3 were obtained from the samples obtained from the DEAE-52 cellulose column after defatted and deproteinized.The molecular weight distribution,the basic form of appearance,Congo red and anti-oxidation in vitro assays were studied.The results showed that:the molecular weight of the three components of the dispersion coefficient was relatively concentrated,respectively,1.4,1.9 and 1.4.The morphology of the surface was analyzed by SEM scanning,which was fibrous,patchy or debris,Congo red analysis showed that the polysaccharides without three helix structure,in aqueous solution was a random coil chain conformation.The antioxidant results showed that polysaccharide had a better DPPH radical scavenging ability and their half inhibition rate IC50values were 3.14,2.85 and 2.31 mg/mL,respectively.And they contained the ability of scavenging free radicals ABTS+,which PS-D3 equivalent antioxidant capacity(TEAC)was the best of 53.83 μmol Trolox/g sample.The study of the basic physical and chemical properties of the polysaccharide from the Pueraria provided the theoretical basis for the development and comprehensive utilization of the Pueraria.
