中文摘要: 采用单因素试验设计、正交试验设计法对山核桃外蒲壳中胡桃醌提取进行了优化,获得了料液比、提取温度、提取时间等参数,并建立了参数方程。由试验结果可知,采用超声波提取法、直接浸提粉末样品,甲醇提取率最高,考虑到安全问题选择乙醇为提取溶剂,提取溶剂乙醇效果好于甲醇和石油醚,且95%乙醇效果好于50%和70%乙醇,料液比以1∶40(g/mL)、提取时间和温度分别为50 min和40℃提取效果较佳。正交试验结果表明最佳工艺为B2A3C3,即料液比1∶40(g/mL),提取时间50min,提取温度50℃时,提取量为0.0625mg/mL,是非正交试验结果平均值的1.3倍。
Abstract:Thesinglefactorandorthogonalexperimentdesignmethodswereutilizedtooptimizetheextractioncon ditions of juglone from Hickory Hull.The results showed that the ultrasonic extraction method,methanol as extraction reagent had higher extraction rate,but for safety consideration,ethanol was better than methanol and petroleumether,and95%ethanolisbetterthan50%and70%ethanol,the material liquid ratio of 1 ∶40(g/mL),extraction time and temperature of 50 min and 40℃ showed higher extraction rate.Orthogonal experiment results showed that the optimal conditions of B2A3C3,namely the material liquid ratio of 1 ∶40(g/mL),extraction time and temperature of 50 min and 50℃had the optimal extraction rate of 0.062 5 mg/mL,which was 1.3 times than the average value of single factor experiments.
