中文摘要: 以荧光微球作为新型标记物,采用EDC/NHS两步法偶联,制备荧光微球链霉亲合素复合物。复合物颗粒均一、性质稳定。以其作为探针,建立检测牛奶中生物素含量的荧光免疫层析法。本方法特异性良好,最低检出限低至3 ng/mL,线性范围5 ng/mL~100 ng/mL,回收率90%~110%,精密度CV<10%;与微生物法对比,测值相关系数为0.975。
Abstract:A composition with a character and uniform size which contained fluorescentmicrospheres as a novel lable was obtained by an EDC/NHS two-step method.The biotin in milk was detected by the immunofluorescence chromatography which had this compositoin as a probe.The method showed good specificity with the detection limitof3 ng/mL,the linear range of 5 ng/mL-100 ng/mL,the recovery of90%-110%and the precision CV less than10%;the correlation coefficient of measuring value was 0.975 compared with microbiological method.
