柱前衍生RP-HPLC法测定仙鹤草中氨基酸含量

中文摘要: 采用柱前衍生RP-HPLC法对仙鹤草中游离及水解氨基酸进行测定。以异硫氰酸苯酯为柱前衍生化试剂。以异硫氰酸苯酯为柱前衍生化试剂;色谱柱采用Ultimate Amino Acid氨基酸柱(4.6 mm×250 mm,5μm);流动相A为80%(体积分数)的乙腈-水溶液,流动相B为7%(体积分数)的乙腈-CH3COONa溶液(pH=6.5);检测波长为254 nm。21种氨基酸在0.001 9μmol/mL~2.700μmol/mL内呈良好的线性关系(R2>0.999 0)。21种游离氨基酸平均回收率在95.84%~ 111.34%之间,RSD在1.23%~2.54%之间(n=6);水解氨基酸平均回收率在90.79%~111.23%之间,RSD在1.29%~ 2.21%之间(n=6)。柱前衍生RP-HPLC法灵敏、准确,具有良好的重复性和稳定性,适合仙鹤草中氨基酸含量测定。
Abstract:To determine the content amino acid in Agrimonia pilosa Ledeb.by pre-column derivatization RPHPLC.The samples were derived with phenylisothiocy anate(PITC).The column was Ultimate Amino Acid of specific column(4.6 mm×250 mm,5μm);Mobile phase:A was mixture ofacetonitrile-water(80%,volume fraction),B was a buffer solution ofCH3COONa(pH6.5)-acetonitrile(7%,volume fraction),eluting with a gradi ent.UV wavelength was setat254 nm.The linearrange of21 amino acids was 0.001 9μmol/mL-2.700 0μmol/mL (R2>0.999 0);the average recovery rates of21 free amino were ranged from 95.84%to 111.34%(RSD=1.23%-2.54%,n=6),the average recovery ofrates of21 totalamino acids was ranged from 90.79%to 111.23%(RSD= 1.29%-2.21%,n=6).This method was flexible,accurate and repeatable and stable for the determination of amino acids in Agrimonia pilosa Ledeb..
