微波辅助提取金钗石斛多糖及体外抗氧化研究

中文摘要: 采用微波辅助提取金钗石斛多糖,通过单因素正交试验法考察料液比、提取功率、粉碎程度和辐射时间对金钗 石斛多糖提取率的影响,确定其最佳提取条件;通过金钗石斛多糖对 ABTS+自由基清除作用测定其体外抗氧化活性。结 果表明:微波辅助提取金钗石斛多糖的最佳提取工艺条件为:料液比 1 ∶ 20(g/mL),提取功率为 400 W,粉碎程度 60 目, 辐射时间为 30 min,在此条件下多糖得率为 3.16 %。金钗石斛精制多糖对 ABTS+自由基的清除率可达 81.9 %,表明其具 有良好体外抗氧化作用。
Abstract:Crude polysaccharides were extracted from Dendrobium nobile Lindl. by microwave-assisted. Opti- mal extraction conditions were explored to evaluate the effects of solvent to material ratio, extraction power, material diameter and extraction time on extraction rate of polysaccharides through single factor orthogonal test method. Dendrobium nobile Lindl. polysaccharides in vitro antioxidant activity were measured by its scavenging effect on ABTS+ free radical. The results showed that the optimal microwave-assisted extraction conditions of polysaccharides from Dendrobium nobile Lindl. were the material-liquid ratio of 1 ∶ 20 (g/mL), the extraction power of 400 W, the extraction material diameter of 60 mesh and the extraction time of 30 min. Under the opti- mal extraction conditions, the extraction rate of polysaccharides was up to 3.16 %. Dendrobium nobile Lindl. re- fined polysaccharides on the clearance rate of ABTS+ free radical could reach 81.9 %.Therefore, the polysaccha- rides from Dendrobium nobile Lindl. have strong antioxidant activity in vitro.
