中文摘要: 建立了同时检测曲霉菌代谢物中黄曲霉毒素和同系物的高效液相色谱-线性离子阱质谱测定方法。 产黄曲霉 毒素 B1(AFB1)寄生曲霉(菌株 3.124)经 PDA 固体培养基培养,提取净化后经线性离子阱(QTrap)质量分析器分析(正离 子模式,多反应检测),以 AFB1 二级碎片(MS2)信息和其同系物分子式信息预测代谢物 MRM 方法,检出 3.124 代谢物中 黄曲霉毒素 B2(AFB2)、黄曲霉毒素 G1(AFG1)、黄曲霉毒素 G2(AFG2)、O-甲基杂色曲霉素(MST)、杂色曲霉素(ST)5 种真 菌毒素,并用标准品进行了确证。结果表明,AFB2 和 AFG2 在 0.5 μg/L~40 μg/L,其它 4 种代谢物在 0.2 μg/L~40 μg/L 范围 内线型关系良好,相关系数均大于 0.99。本方法 6 种代谢物日内回收率 81.3 %~92.3 %,相对标准偏差(RSD)为 3.4 %~6.2 %; 日间回收率 82.1 %~91.7 %, RSD 为 3.8 %~7.7 %。该方法为快速检测霉菌代谢物中的同系物提供新的方法。
Abstract:A stimulate method for determination of aflatoxin and aflatoxin -like compounds in Aspergilli metabolite was developed by high performance liquid chromatography- linear ion trap mass spectrometry. A. parasiticus (strain 3.124) which produce AFB1 was cultivated by PDA solid medium, then the metabolites of strain 3.124 was analyzed by mass analyzer of QTrap(positive ion mode, multiple reaction monitoring(MRM)) after extraction and purification, and found five mycotoxins of AFB2、AFG1、AFG2、MST and ST through the MRM methods predicted by MS2 of AFB1 and structural formula of aflatoxin-like compounds, and was confirmed by standard. The linear range of AFB2 and AFG2 was 0.5 μg/L-40 μg/L, the other 4 metabolites showed good linearity in the range of 0.2 μg/L-40 μg/L with the correlation coefficients more than 0.99. The recoveries of in- ter-day ranged from 81.3 % to 92.3 % with the relative standard deviations of 3.4 %-6.2 %, The recoveries of intra-day ranged from 82.1 % to 91.7 % with the relative standard deviations of 3.8 %-7.7 %. A new method for rapid determination of aflatoxin and aflatoxin-like compounds in Aspergilli metabolite was provided.
