中文摘要: 探讨基于代谢组学法考察金银花醇提物对DMN诱导的大鼠急性肝纤维化损伤的保护作用。建立DMN诱导的大鼠急性肝纤维化损伤模型,采用DMF衍生大鼠尿液,用气相-质谱的代谢组学方法,从整体代谢水平上考察尿液中内源性小分子代谢物的变化,使用主成分分析(PCA)法探寻大鼠尿样的代谢物谱的模式变化及潜在的生物标记物。尿液的GC-MS指纹图谱可知DMN所致大鼠尿液中内源性小分子代谢物中有3种物质含量增加和6种物质含量降低,发生了代谢网络扰动象。经金银花醇提物干预后,其尿液代谢表型有向正常范围回归的趋势,呈现代谢网络修复的结果。说明预先灌胃金银花醇提物对DMN染毒大鼠的生理及代谢状况产生了有效的保护作用。
Abstract:Discussed the effect of protection on the liver fibrosis induced by DMN in rats base on the method of metabolomic. The rats model was established by DMN with acute injury of liver fibrosis and all the urine were derived with DMF. Then investigated the endogenous small molecule metabolites in urine from the whole lever of metabolism changes. Researched the spectral pattern change of metabolites and potential biomarkers in rat urine with principal component analysis(PCA)method. It found that the peak areas of 6 metabolites significantly decreased and 3 metabolites significantly increased after DMN injection and the variations were inhibited after LJE intervention. The metabolic disorders have been significant adjustments and the urinary metabolic phenotype return to normal trend. Based on these results,LJE have protection on liver.
