中文摘要: 主要研究鱼头多糖对K562细胞的抑制作用。通过MTT法检测鱼头多糖对K562细胞的增殖抑制率,HE染色法观察K562细胞形态学的变化,流式细胞仪检测K562细胞的凋亡情况。结果表明,MTT法测定在药物浓度≤200μg/mL时,鱼头多糖对K562细胞增殖有明显抑制作用,其抑制率与时间、浓度呈正相关。HE染色显示多糖作用于K562细胞后出现了明显的凋亡和坏死同时发生的现象,且表现出时间依赖性。流式细胞仪检测多糖对K562细胞周期分布有显著影响,且凋亡现象明显,并伴有明显的坏死现象。在多糖作用72 h后,凋亡细胞的比例高达细胞21.28 %。因此,提示鱼头多糖对K562细胞的增殖抑制作用明显。

Abstract:The aim of this paper was to study the effect of human leukemia K562 cells treated by the fish head polysaccharides. MTT method was used to test the proliferation inhibition rate of K562 cells,and HE staining was used to observe the morphological changes of K562 cells,and flow cytometry was used to test the apoptosis of the K562 cells. The result indicated the proliferation of K562 cells was obviously inhibited,and the inhibitory rate was positive correlation with time and concentration when the drug concentration was less than 200 μg/mL. HE staining displayed the K562 cells presented obvious apoptosis and necrosis phenomenon with time dependence. The flow cytometry testing indicated the polysaccharides had a significant impact on the K562 cell cycle distribution,as well as obvious apoptosis and necrosis phenomenon. The rate of apoptosis cells run up to 21.28 %. Therefore,it showed the fish head polysaccharides had obvious impact on the proliferation inhibition of the K562 cells.
