中文摘要: 抗菌肽是一类免疫效应因子,它在生物机体的先天免疫系统中起到重要的作用,具有广谱抗菌、抗寄生虫、抗病毒、抗癌、抗氧化等多种生物学活性,尤其是其对某些癌细胞具有高毒性,对正常细胞低毒性的特性引起了研究者们广泛关注。以中华圆田螺作为研究对象,初步提取抗菌肽,测定其抗菌效果、理化特性和抑癌活性。结果表明:粗提液抑菌效果明显;其抑菌活性随着处理温度的升高,处理时间的延长呈现逐步降低的趋势,但仍表现出较好热稳定性;在弱酸性(pH值4~6)条件下,随着酸性的降低抑菌活性呈现增加的趋势,在中性和碱性(pH值7~12)条件下,随着碱性的增加抑菌活性不断降低;随着冻融次数的增加,抑菌活性略有变化;粗提液对A549细胞的增殖表现出抑制作用。
Abstract:Antibacterial peptide is a kind of immune effect factor, it plays an important role in innate immune system of biological organism. It has many kinds of biological activity, like broad spectrum of antibacterial, resisting parasites, antivirus, antitumor, antioxidation and so on, especially its high toxicity to some kinds of cancer cells and low toxicity to normal cells cause the researchers’ attention. Cipangopaludina cathayensis is used as the research object, this study extracts antibacterial peptide preliminary from Cipangopaludina cathayensis and determines its antibacterial effects, physical and chemical properties and tumor-suppressor activity. The results show that: the bacteriostatic effect of the crude extraction is obvious; with the treating temperature increasing, treatment time extending, antimicrobial activity reduce gradually, but still show good thermal stability; Under the weak acidity (pH 4~6)conditions, antibacterial activity tended to increase as the acidity reduces. Under the neutral and alkaline (pH 7~12) conditions, antibacterial activity reduces as the alkalinity increases; With freezing and thawing times increasing, antibacterial activity changes slightly; crude extraction shows inhibition to A549 cells’ proliferation .
